CD77, also known as Globotriaosylceramide (Gb3), Burkitt Lymphoma-Associated Antigen (BLA), Ceramide Trihexoside (CTH), and the Pk Blood Group Antigen, is a key glycosphingolipid with diverse biological roles. Primarily expressed on certain germinal center B lymphocyte subsets, especially centroblasts, CD77 is abundant in tonsils and mucosal lymphoid tissues, but relatively scarce in peripheral blood, bone marrow, and the spleen.
Notably, CD77 acts as the cellular receptor for Shiga toxin, a bacterial toxin produced by Shigella dysenteriae and specific strains of Escherichia coli. Furthermore, studies indicate that crosslinking CD77 can trigger B-cell apoptosis, suggesting a potential role in immune regulation and programmed cell death.

Clinical Significance of CD77
CD77 is highly expressed in various malignancies, notably Burkitt lymphoma (BL) and head and neck cancer (HNC), contributing to tumor proliferation and metastasis. Additionally, it is associated with Fabry disease (FD), a genetic lysosomal storage disorder caused by α-galactosidase A deficiency, leading to Gb3 accumulation. The therapeutic agent migalastat enhances enzymatic function, reducing Gb3 deposition and alleviating symptoms.
Moreover, CD77 plays a critical role in Shiga toxin-induced cytotoxicity by activating calcium ion (Ca²⁺), cyclic AMP (cAMP), and ceramide signaling pathways, ultimately inducing cell apoptosis and severe pathological reactions.
